There are more reasons, than it just being adorable, for serving a signature cocktail at your wedding. For one, your caterer and bartenders can prepare the concoction in advance. Servers can stand ready as guests arrive so that they are simultaneously greeted and quickly refreshed. Then as cocktail hour unfolds, servers can roam around with trays of the drink which will also reduce the wait at the bar.

Choose a cocktail that you both like or that is sentimental, and work with your caterer to make sure it's something in their repertoire. Design a little tray tent or custom cocktail napkins announcing the drink and choose a name that celebrates you both (Sue + Matt = MattSuetini), your locale (Boston Bomber, Waltham Wallop, Provincetown Punch) or something to note the occasion (Wedded Bliss, Always & Forever, The Ball & Chain). Have fun; a signature cocktail should be light, fun and bring a smile to your guests' faces.
